Do i add the whole bottle if im just changing the rear or half in front n rear

The whole 4 ounces goes in the rear differential along with the recommended weight gear oil on Mopar vehicles with the limited slip differential in the rear. Unless you have an old model jeep with a low bias limited slip differential in the front axle or have swapped in an aftermarket lmited slip in the front axle this fluid shouldnt need to be added to the front axle and even at that you would need to consult the factory recommendations for your vehicle concerning the front axle. I only made the change on my 2 wheel drive Dodge Ram 1500 hemi Sport and it called for the whole 4 ounces of limited slip differential additive in the rear with 75w-140 gear oil.

I'm getting an aftermarket cover for my 08 Ram, with the new cover it'll hold about 4 quarts of diff fluid. Will I need 2 bottles of addtive?

Unless the after market cover expands the fluid capacity of your diff (which wouldn't make any sense) the one bottle should cover it. Otherwise look up the specs and do the math.
